At a Glance
Researchers at Colorado State University have developed a series of DNA plasmids based on the pTE3’2J Sindbis virus plasmid that can be transfected into mammalian cells to produce non-infectious Sindbis virus replicons (viral genomes with structural proteins removed, non-infectious) and fully-infectious Sindbis viruses. These replicons and viruses can be used to study a wide range of cellular processes in mammalian and insect cells, express various recombinant proteins in these cells, or infect mosquitoes or other species to study infection or their physiology.

Replicon Producing Plasmids
pBG60 Recombinant Sindbis Replicon Expression Plasmid – Gateway Compatible
Recombinant plasmid pBG60 containing Sindbis virus (SINV) that allows for fast gene of interest addition with Gateway recombination.
Highlights:
- Contains a Gateway recombination cassette for rapid addition of genes of interest into the replicon 3’ subgenomic promoter
- Expresses Blasticidin-S-deaminase (BSD) protein for selection
- Can be used in poorly or non transfectable cells when co-transfected with the packaging plasmid pBG256
pBG68 Recombinant Sindbis Replicon Expression Plasmid
Recombinant plasmid pBG68 produces Sindbis virus (SINV) replicons in cells that allows for fast and easy expression of your gene of interest.
Highlights:
- Can be transfected into mammalian cells to produce replicating but non-infectious Sindbis virus genomes
- Expresses Blasticidin-S-deaminase (BSD) protein for selection
- Can be used in poorly transfectable cells when co-transfected with the packaging plasmid pBG256
pBG78 Recombinant Sindbis Replicon Plasmid – GFP Expressing
Recombinant plasmid pBG78 produces Sindbis virus (SINV) replicons in cells that can be easily visualized via GFP expression.
Highlights:
- Can be transfected into mammalian cells to produce replicating but non-infectious Sindbis virus genomes
- Expresses GFP for easy visualization of viral genome replication in live cells
- Expresses Blasticidin-S-deaminase (BSD) and Green Fluorescent Protein (GFP)
- Can be used in poorly transfectable cells when co-transfected with the packaging plasmid pBG256
pBG256 Recombinant Sindbis Virus Replicon Packaging Plasmid
Recombinant packaging plasmid pBG256 lacking the viral nsP1-nsP4 replication genes and can be used to combine plasmids with pBG60, pBG68, pBG78.
Infectious Virus Producing Plasmids
pBG167 Recombinant Sindbis Virus Expression Plasmid
Recombinant plasmid pBG167 expressed infectious Sindbis Virus (SINV) and allows for fast and easy expression of your gene of interest.
Highlights:
- Plasmid can be transfected into mammalian cells to produce infectious Sindbis virus
- Unique XbaI site behind 3’ subgenomic promoter can be used to express genes of interest during infection
pBG210 Recombinant Sindbis Virus Expression Plasmid – Gateway Compatible
Recombinant plasmid pBG210 containing Sindbis Virus (SINV) allows for fast gene of interest addition with Gateway recombination.
Highlights:
- Contains a Gateway recombination cassette to allow for efficient addition of genes of interest behind 3’ subgenomic promoter
pBG218 Recombinant Sindbis Virus-GFP Expression Plasmid
Recombinant plasmid pBG218 expresses Sindbis virus (SINV) expressing GFP during viral replication for easy visualization.
Highlights:
